MacBook Pro not starting up!

When I try to turn on my Mac I just get a grey screen with a for folder in the middle, with a question mark on it. Any ideas?!?!

Try booting up holding the Option key, then using the arrows on the keyboard, select your internal drive & press return

By this do you mean nothing was displayed at all in Startup Manager Mode? If you have a later OS installed you could try Recovery Mode (if it showed up) then choose Disc Utility

Also anything else we need to know regarding the events leading up to this?

If you have a Software Install Disc you could boot from it & go into Disc Uitility in the Menu & Verify Disc.
