iMac won't Start up

When we switch on our iMac, we just get a grey screen with a No Entry sign, or get the below recurring error message. We've tried starting up in Safe mode and disconnecting the peripheral devices but to no effect. Any help would be appreciated. Thank you.

The no-entry sign usually means a failing hard drive.

Try booting up holding the Option key after the Startup chime and go into Disk Utility and Verify Disk.
